摘 要:
脱氢抗坏血酸还原酶是抗坏血酸代谢循环中的关键酶, 在多种植物中与抗胁迫相关。为了获得抗盐碱植物星星草中 该基因序列, 利用 RACE 技术, 从星星草中克隆出脱氢抗坏血酸还原酶基因(PtDHAR)的 cDNA 全长序列, 其 GenBank 登录 号为 HM125046。PtDHAR cDNA 核苷酸序列长度为 987 bp, 开放阅读框为 639 bp, 编码 213 个氨基酸。该基因编码的氨基 酸序列与水稻、小麦等禾本科作物具有很高的同源性。Northern 杂交分析表明, 该基因在盐碱胁迫下表达量显著升高。关键词:星星草; 脱氢抗坏血酸还原酶; 基因克隆; 表达分析

Abstract:
Dehydroascorbate reductase (DHAR) is the key enzyme in ascorbate metabolic cycle which was reported related with stress resistant in many plants. RACE technology was employed to obtain full-length cDNA from Puccinellia tenuiflora named PtDHAR (GenBank accession number HM125046), which was 987 bp length. The open reading frame encoding 213 amino acids which had a high identity with DHAR from Oryza sativa and Triticum aestivum. Northern blotting analysis showed that the expression level of PtDHAR under saline-alkali stress was significantly up-regulated than that with no stress.Key words: Puccinellia tenuiflora; dehydroascorbate reductase; gene clone; expression analysis
